    Chloroquine ineffective on exoerythrocytic

Chloroquine has been extensively used in mass drug administrations, which may have contributed to the emergence and spread of resistance. It is recommended to check if chloroquine is still effective in the region prior to using it.

Chloroquine is a 4-aminoquinoline with antimalarial, anti-inflammatory, and potential chemosensitization and radiosensitization activities. Although the mechanism is not well understood, chloroquine is shown to inhibit the parasitic enzyme heme polymerase that converts the toxic heme into non-toxic hemazoin, thereby resulting in the accumulation of toxic heme within the parasite. Chloroquine concentrates in the food vacuole up to 1000-fold. Chloroquine, exists in unprotonated form, CQ, monoprotonated form, CQ + and diprotonated form, CQ ++ form 13. Unprotonated form of chloroquine is membrane permeable and it freely diffuses into the red blood cell. It then continues to diffuse into the DV. Quinacrine and quinine as used against P. cathemerium in the canary Kikuth and Mudrow, 1939 and against P. gallinaceum in the chick Coggeshall, Porter and Laird, 1944 have been ineffective against the exoerythrocytic stages.

The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

Chloroquine Aralen is used for preventing and treating malaria and amebiasis, an infection of the intestines caused by a parasite. Chloroquine Aralen - Side Effects, Dosage, Interactions. Chloroquine is not active against the gametocytes and the exoerythrocytic forms including the hypnozoite stage P. vivax and P. ovale of the Plasmodium parasites. In vitro studies with Chloroquine demonstrated that it is active against the trophozoites of Entamoeba histolytica. Extracorporeal elimination techniques are ineffective. Arrhythmias are best treated by correction of hypoxia and electrolyte disturbances, and sodium bicarbonate to correct metabolic acidosis. Hypokalaemia resulting from intracellular potassium transfer is closely related to severity of chloroquine poisoning and should be corrected cautiously.

Plaquenil, Colchicine & Antihistamine? Lupus Forums at. I take antihistamines and Plaquenil plus many other meds, not sure about the colchicine. One thing to remember is that some medications we may currently take can perhaps make us drowsy, antihistamines on top may add to that. Antihistamine may well help with the itch after showering.

PLAQUENIL" Rheumatoid Arthritis Community - Support Group I have noticed an improvement but it's very little. I have 2 days a week, after my MTX, where I'm just sick. I can't do anything. I've been on the MTX now for about a month and when I see my rheumy in 2 weeks I'm going to see if I can try something else or stop taking it. I don't know if the plaq or MTX or prednisone is what's helping.
